| I. Background | |
| Notch1 作為 Notch 受體家族核心成員，由胞外配體結合域（EGF 重復序列）、跨膜域 及胞內域（NICD）組成。當 DLL4 結合 Notch1 胞外域后，引發(fā)兩步蛋白酶切割：ADAM 蛋白酶（如 ADAM10/17）切割胞外段，隨后γ-分泌酶切割跨膜區，釋放 NICD 進(jìn)入細胞核。 NICD 與轉錄因子 CSL 及輔激活因子 MAML 形成復合體，激活下游靶基因（如 Hes1、 Hey1、Myc），調控干細胞維持、細胞增殖及分化。 DLL4 與 Notch1 是 Notch 信號通路 的關(guān)鍵組分，通過(guò)細胞間接觸依賴(lài)性信號調控發(fā)育、 血管生成及疾病進(jìn)程。DLL4 是 Notch 配體家族成員，作為跨膜蛋白特異性地結合并激 活 Notch1 受體，驅動(dòng)細胞命運決定、組織穩態(tài)及病理重塑。 |
